  • How To Use

    Sample Information
    A sample size of 5 μl should be used. An appropriate sample can be any one of the following:

    • Anticoagulated whole blood (heparin or citrate)
    • Plasma
    • Serum

    IMPORTANT! This test uses a special self-filling pipette to accurately deliver a small sample volume to the test. The pipette will fill automatically by capillary action when placed in the sample. Only squeeze the bulb to deposit the sample on the test.

    1. Grip the pipette by the stem. Hold a tube of sample and pipette at an angle to allow proper filling of pipette.
      • Do not use this kit or any of its components after the expiration date. Use the test within 10 minutes of opening the pouch.
      • Without squeezing, place the tip of the pipette in the sample. The capillary tube in the pipette will fill automatically in 2-3 seconds.
      • Remove pipette from sample when the capillary tube has filled.
    2. Touch the tip of the pipette to the sample pad and carefully squeeze the bulb to express the sample (5 μl), making sure fingers are covering the vent holes on the pipette bulb.
    3. Add 3 drops of buffer solution.
    4. Wait approximately 10 minutes.
    5. Read test results.

    Sample Information

    • WITNESS Lepto detects the primary immune response (immunoglobulin M [IgM] antibodies) which develops within one week of exposure to canine leptospires. WITNESS Lepto was shown to detect four important canine serovars – Canicola, Grippotyphosa, Icterohaemorrhagiae, and Pomona. The WITNESS format is referred to as Rapid ImmunoMigration (RIM™), sometimes referred to as lateral flow or rapid immunochromatography.
    • After the sample and buffer are added to the sample pad (window 1), Leptospira antibodies, if any, in the sample migrate across a nitrocellulose membrane along with the gold-labeled anti-dog IgM antibody (conjugate) and form a Leptospira antibody/colloidal gold-labeled antibody complex. Leptospira antigen binds the Leptospira antibody/colloidal gold-labeled antibody complex at the test line.
      In a positive sample, this generates a pink-to-purple–colored reaction line in (window 2). Excess colloidal gold-labeled antibody across to the control line, where antibody directed to the colloidal antibody binds. This causes formation of a pink-to-purple–control line (window 3).

    A test result should always be interpreted in the context of all available patient history and clinical information.

Precautions

  • Do not use this kit or any of its components after expiration date.
  • Kits should be stored at 35° to 77°F (2° to 25°C). Kit should not be frozen.
  • Use the test within 10 minutes after opening the pouch.
  • Avoid touching or damaging the membrane in the sample well or the results window.
  • The WITNESS device should be placed on a flat, horizontal surface while performing the test.
  • Use a separate pipette for each sample.
  • Ensure that the capillary tube in the sample pipette has been completely filled (an air bubble will be visible at the top of the capillary tube if it has not been completely filled).
  • Hold buffer bottle vertically when dispensing buffer.
  • Handle all samples as biohazardous material.
  • Saline buffer is preserved with sodium azide.
